Suspension cells are hard to transfect, thus making it difficult to overexpress or knockdown a gene. The alternative strategies are either to do electroporation, nucleofection or lentiviral transduction. These days many companies have developed transfection reagents (they claim), which are suitable for overexpression or knocking down of a gene in hard to transfect or suspension cells. I have to silence a gene in acute myeloid leukemia HL-60 cells (a type of suspension culture). Does anyone have personal experience in using such type of reagent for siRNA delivery in suspension cells? Also please mention the specification of the transfection reagent. Thanks.
