I need to know about the problems in formation of somatic embryogenesis and difficulties in establishment of plantlets from callus (indirect organogenesis) in plant tissue culture.
Is there any report that says about the the problems with the formation of indirect organogenesis/ indirect morphogenesis/ somatic embryogenesis in in-vitro cultured plants or any physiological and biochemical molecules responsible for it?.
Are any plants that shows the above said problems?
If there are any reports kindly attach the links if possible. it is one of the demerits of the plant tissue culture probably.

Dear Sachet,
There are merits as well as demerits of the topics u have written. One of the problem is chance to get variant through indirect organogenesis but on the other hand u can get superior somaclone which may have high metabolite content. Somatic embryos are generally true to type in nature but getting them is difficult as it requires 2 different media and optimization takes time.
So overall it's up to aim of your study what is beneficial or not. And there are many reports available in good journal u can search or name for particular plant if I have I will send you.

There are many recalcitrant species that do not readily form embryoids in anther culture. some of them can be induced to form embryoids by some kind of shock: cold, heat, osmotic etcetera. Almost all legumes are recalcitrant, as are certain rice and wheat types.

Dear @Luis Carlos da Silva Ramos ji, thnak you for the suggestions that you mentioned....I am using MS media and WPM for a woody climber so is there any criteria for the plant to give plantlets from in-vitro cultured callus? do anyone reported the physiology behind the problem? does each and every plant is 100% totipotent(according to the practical means) I am asking...
