I am facing a germination problem in the seeds of some weed species, which I am working on, because of seed dormancy. to break the seed dormancy I need to provide gibberellic acid in the MS media, What is the correct way of dissolving the Gibberellin acid and what is the correct method to provide that along with the solid MS media?
Autoclave MS and wait for it to cool down to 50C. Use a water bath to prevent premature gelification. Once the MS is at 50C, add a GA solution using a syringe and a syringe filter (0.2~0.4 µm) if you wish to keep the media sterile.
Using MS to germinate seeds is probably useless... you can do with plain agar and GA.
GA solution: GA can be dissolved in water, but pH will need to be adjusted around 6.5 to ensure it dissolves fine.
