I've been facing a problem LC-MS/MS analysis. I have a protein that in produced in E. coli and it is actually deposited in inclusion body.

Protein contain methionine as a start codon, during LC-MS/MS analysis I could not find any sequence coverage of methionine thats indicating lack of Methionine in N-terminal sequence.

Is there any proven data that cell may expel the N-terminal methionine during synthesis or during during deposition in IB.
