I grow 100,000 cells/2mL media in 35mm glass bottom dish and after 48 hours transfect them with DNA(membrane protein) and will be ready for imaging after 24 hours. I want to increase cholesterol content in cell membrane. I am wondering how to make beta-cyclodextrin: cholesterol complex and when/how /how much to add it into the cells during cell culture.
Maybe one possibility, it is you use this drug diluted in the medium in a controlled and known concentration, when your cells have established itself, for example, in a case of adherent cells, after these cells have adhered in the plate.
In this case, you will need make a dose-response curve and a time-response curve to try to establish how each dose it is supported by your cells, and how these cells act along the time when exposed to the dose that you will choose.
