I am trying to switch from NaCl to KCl in my buffer. As soon as I add 4M KCl solution to DI water and HEPES, it precipitates. Would heating the buffer alleviate the problem? Or should i add KCl to just DI water without any HEPES in it? also the pH was fine: 7.4
Adjust the pH of the HEPES buffer to the desired value in the usual way, using a HEPES solution that is a little more concentrated than needed for the HEPES-KCl mixture, then slowly add the required amount of concentrated KCl while stirring the solution. Finally, bring the volume up with distilled water to the final volume needed to bring the concentration down to the desired value.
Alternatively, prepare separate HEPES (pH-adjusted) and KCl solutions at 2X their final concentrations, then mix them together.
