I am working on cell line establishment from fish. I am using 20% FBS but recently I have read somewhere that FBS 2% is used for attachment of cells in the first day and then 10% FBS is used for the rest of the culture. Do you think that reduced FBS concentration is really effective in attachments of cells? How?
I can tell for sure, you do not need to use 20% FBS. Did you try using 2% FBS, was the attachment okay or better than before? Then, you may follow it as that is cost effective, and the cells are not going to die in a day just because they are in 2% FBS.
I have cultured primary vascular cells from the rat in serum free but plus H12 nutrient mixtures.

According to the experience and studies, some fish primary cells are sensitive to level of FBS. For example in fish macrophages culture, you should use low serum concentration for the adherence step, (eg 1%). In high serum (5-10%) macrophages do not adhere as well.
