Or can I use DMEM with phenol red? Does the presence of phenol red interfere in the determination of NO?

Not necessarily.If u feel the colour of your media and griess product can misinterpret, i would suggest you to go for a preplate reading with your supernatant before adding greiss reagent thereby u can avoid errors.
