I am working with mouse blood and bone marrow samples and enumerating immune cells.
As I understand, an anti CD16/CD32 is used as an Fc block for mouse samples.
However, I am not sure whether CD16/CD32 is a marker for any specific cell type. So can I use it to enumerate a specific cell type or am I simply blocking by adding this antibody to my sample?
In humans CD16/CD56 is a marker for NK cells. Is it the same in mice also?

Mouse CD16/CD32 epitope is expressed on B cells, monocytes/macrophages, NK cells and neutrophils, so has a wider cell distribution than the human CD16/CD32 epitope.
I think its an IGF receptor,should be found on splenocytes.

B cell expresses only CD32. Monocytes/macrophages, NK cells and neutrophils express both CD16 and CD32. No monoclonal antibody specific for mouse CD32 is commercially available so far.
CD16 and CD32 are Fc gamma receptors (FcyRIII and FcyRII respectively), that bind IgG with different affinities. Eg for CD32 typical immune complexes are needed for downstream responses.
The manuscript Charan Thej is referring to describes the Cd32B isoform, which is an inhibitory form, in contrast to CD32a which is activating.
CD32 is FCGR2 and CD16 is FCGR3.
- CD32 is expressed in a variety of cells and modulates B cell activation, Dendritic cell and immune cells (mast cells, neutrophils and macrophages),
- CD16 is expressed in a variety of cells too and activates lck in NK cells, and lyn and hck in monocytic and mast cells.
This receptors are also involved in complement cascade.
With such a wide expression CD16/32 cannot be used as a cell marker in mouse.

Both CD16 and CD32 are Fcγ receptors for the IgG antibody's Fc region. CD16 is FcγRIII while CD32 is FcγRII. These are expressed on a variety of cells including B cells, monocytes/macrophages, NK cells and neutrophils. Fc blocking is usually done by using their respective antibodies!
