A lot of people use lambda phage DNA as stuffer DNA for a variety of applications. I was wondering if anyone knows if there would be any expression of the phage genes if the stuffer is used in eukaryotic cells? Do phage promoters work in mammalian cells?

Assuming that you meant whether the lambda bacteriophage genes are expressed in the eukaryotic cells, a simple answer is NO. It is known that bacteriphage gene regulatory regions ("promoter" sequence) do not efficiently bind to eukaryotic transcriptional regulator proteins and do not support "transcription".

totally agree with Divaker, and furthermore add that plain DNA from lambda can not get inside eukaryotic cells. Lambda phage needs its coat proteins to "inject" lambda DNA inside E. coli, and I don't think that it can do the same with an eukaryotic cell
