Hello, I bought Amphotericin B from a company. Their instruction is to dissolve it in DMSO at a concentration of 3 mg/mL. Is it suitable to add 10 microliter of this solution (ie 6 micro gram of amphotericin B) to a flask having 5 mL of culture media?
it is unadvisable to add any antifungal to cell culture .
ok it will eradicate fungal infections but it has a nasty effect on cells response (especially T cells which i work on)
I agree that most anti-mycotics tend to be hard on animal cells, but there are situations where it necessary. The issue with DMSO is that it is hypertonic [3.75M osmolarity] compared to saline [.15M osmolarity]. But the volume you are suggesting 10 microliters into 5 ml medium should be in the tolerable range for live cells. The question is why do you want to do this and how long to you want to keep the amphotericin B in the culture?
