Is it possible to isolate PBMC with Ficoll from frozen blood ?

I tried to isolate PBMC from frozen blood, but nothing happened. Do you know why? Is it because all the cells exploded?

Yes, it is very lickely that the PBMCs were not viable after thawing due to cell lysis. If you freeze cells (here blood cells) without a proper cryoprotectant, ice crystals form inside the cells, causing cell damage and cell lysis upon thawing. PBMCs should always be isolated before freezing.
