Hi,
I'm currently doing a CARD-FISH method on soils samples to detect methanogens and methanotrophs communities.
For my first steps, I need to wash my samples 2 times with 1x PBS to remove the paraformaldehyd and stop cell fixation.
The protocol that i'm following does not explain in detail how to wash the samples but only specify to centifuge after each washing.
My samples are in 2mL tests tubes containing 0.5 cm3 of soil, 320 uL of 4% paraformaldehyd and filled with 1x PBS.
Can anyone explain to me how to wash a sample ?
