Hi Sean. I use to purify neutrophils from bone marrow,but I would like tô get immature neutrophils and mature them in vitro. Do You Know If is It possible?

Hi, so did anyone of you tried that protocol for isolation of neutrophils? I'm trying to use it but can't get it right.
This is what I get (photo). I get a clear halo between what I think is the media and the first histopaque layer, and then between the 2 layers of Histopaque, I don't see nothing (where neutrophils are supposed to be). Do you have any idea why this could be happening? Thanks!
Isolate neutrophils from bone marrow by percoll or similar gradient did not work to me. Sometimes I got it but not always. I prefere isolate them by magnetic beads. Have you ever try it?
