I know that 293-Freestyle cells are a human cell line that can be cultured in shaker flasks. I am interested in growing a moderately large quantity of THP-1 monocytes, but I am a bit short on incubator space. Would THP-1s or other suspension cells survive being shaken? Also, does anyone expect their transcriptional profiles to be terribly altered? (I kind of expect the shear force to alter gene expression) Since monocytes circulate around the body I wonder if they would be resilient enough to survive even gentle shaking.
Any (constructive) input is appreciated! I don't know if anyone has experience with similar cells in a shaken environment. Thanks!
i can answer in the regard of survival
the gentle shaking shouldn't reduce your cells viability primary immune cells and cell lines are often maintained under gentle shaking.

There are some protocols where people have put THP-1 cells under gentle shaking for 3 hrs or for short intervals but culturing the cells for entire time in shaking culture flask, seriously the answer is just to try it by yourself. So you can start by a small cell volume and standardize it by urself if it suits your experiment requirements. Just to be out of the box, I can only recommend that you can culture the cells in a little large amount of media than usually recommended so that the shock effect due to shaking can be minimized. Thats a little bit physics but making the cells shockproof may work for your conditions.
