One of the research groups has shown that they are not able to detect ERK5 protein expression (taking into consideration proper negative and positive control) in brain using immunohistochemistry and they concluded that ERK5 is not present in adult brain. They didn't try western blot analysis to confirm the expression profile. While, I am able to detect ERK5 protein expression in brain tissue using western blot analysis. We have the same animal species and strain. The only difference is the primary antibody from a different company.
Is it possible that one can detect desired protein using western blot but not using immunocytochemistry/immunohistochemistry?
