I have some HL60 and U937 cells I'm growing in 50mL of media in T75 flasks, and I was wondering if it would be okay to do a 1:2 split by just adding 50mL more media to the same flask instead of transferring the cells to two new ones. If it makes a difference, I'm going to be immunoprecipitating and eluting the proteins from them for proteomic analysis (mass spec).

There is no problem if you go for routine subculture and all. But as you mentioned you want to study immunoprecipitation followed by massspec, so better try to spin down the cell suspension at 200 x g for 4 minutes to remove debris and dead cells.

I use to do this, with u397 insect cells, they were in spinning culture flasks. But your method should work, and save money too. Nice.
