Dear all,
I constructed a CRISPR cell line and found that aside from a one bp deletion at DSB, there are also some point mutations far away from the DSB (and a long deletion about 300bp away). The farthest one is 267 bp from the DSB and they are not SNPs. Is this phenomenon normal? I have concern that other people will question the off-target possibility if so.

- High frequency of off target activity is possible with RNA guided endonucleases. Therefore, it is better to use multiple different guide RNAs and then choose the one which gives the least off target effects.
