I need to use tamoxifen during embryonic day 11.5-13.5 in pregnancy mice, even though I use tamoxifen in low doses of 10mg/kg i.p. every day, the female mice were fragile. After c-section, pups were too weak to live more than 4 days.
While finding references about how to use tamoxifen in pregnancy mice, only one paper indicated that using progesterone along with tamoxifen can decrease the implication of tamoxifen.
Did anyone use progesterone in pregnancy mice, and can you share the protocol about it? Thank you very much
I'm encountering a similar issue with my study where I'm administering 3-4 mg/kg of tamoxifen via orogastric gavage to pregnant mice, and I'm experiencing similar difficulties with the health of the mothers and viability of the pups. Some evidences suggest that using progesterone along with tamoxifen can decrease the adverse effects of tamoxifen during pregnancy in mice. One study that explores this is “Progesterone attenuates adverse effects of tamoxifen and estradiol in estrogen receptor positive mammary glands of ovariectomized pregnant mice” by Lee et al., PMID: 27655440. This study provides insights into the protocol for using progesterone alongside tamoxifen to mitigate its adverse effects during pregnancy in mice.
