I would like to increase my virus titer by using hek cell line. I saw somewhere that they decrease the temp of culture after infection. Is there anybody who has such kind of experiment?

Yes, decrease in temperature of culture after infection will help.
The virulence of a microorganism is dependent on the temperature sensitivity of their replication. Temperature sensitivity is the property of a virus to replicate well at the lower temperatures. Adenovirus is one of the respiratory viruses which replicate best at a temperature between 33- and 35-degree C.
At the same time, you should also keep in mind that HEK293 cellular attachment is also temperature dependent. Reducing the temperature of HEK 293 cultures to 30°C may result in up to 60% loss of the cells from the monolayer.
