May I know the different if we measure the cytokine level using ELISA and protein expression (Western blot and RT-PCR)? Some of the journal measure both.
Dear Muhd, It is depend on your propose. You can check expression at two levels and two approaches.
1) Expression at gene level (mRNA) and qualitative approach (e.g. RT-PCR)
2) Expression at gene level (mRNA) and quantitative approach (e.g. Real Time PCR)
3) Expression at protein level and qualitative approach (e.g. Western Blot)
4) Expression at protein level and quantitative approach (e.g. ELISA)
Now based on your propose in your research you can choose one or more above options.

Selam Hanis. As already said it depends on what the purpose is. If the purpose is to quantitate the amount of protein, then ELISA is enough. But if there is interest to not only measure the amount of protein but also identify which cells expressed it then immunocytochemical/immunohistochemical staining followed by/without western blot for confirmation will be necessary. So depending on the purpose, the different techniques of ELISA, ICC/IHC, WB, RT-PCR... may appear in one research paper.
