I see some of my friends filter their DMEM, PBS, trypsin for cell culture use. Can it depend on the type of cell? In witch situations must these be filtered? Anybody know?
If you are ordering ready-made liquid media from companies then you don't need to filter it. The same applies for PBS and Trypsin. However if you are making your media and trypsin using powdered solutions, then it is always preferred to be filtered. In case of PBS, if you are dissolving the salts yourself then you can just autoclave it and filtration is not required.
I also know some labs where people filter the media. Usually, it is labs shared by many different persons, and some may have doubts about the sterile conditions applied by their colleagues... Filtering ready-to-use, sterile complete medium is then a way to ensure that your medium is kept sterile, whoever worked with it before.
