I did ethanolic extraction on my red seaweed using a soxhlet extractor. I'm planning to only reduce my extract from 250 mL to 60 mL. Then, I'm going to dilute it with 40 mL ethanol to achieve 60% of the extract. However, base on my readings, extracts are reduced to dried crude extract before doing dilution. Should I continue my plan or do I really need to dry it first? I really need help. Thank you very much!

You should discuss this with your supervisor in the lab.
The way to reduce to dryness first is used:
- in order to increase concentration of metabolites in dilutions
- to have a better stability for storage
- to be able to add additional steps for purification, e.g. in other solvents
If you know, that conc. of your compounds of interest is high enough to be well above LOQ in analysis and if you do analysis straight after extraction (no storage for more than several days even in the frige) you can try your idea.
