I suggest you try the combination of CpG & CD40L along w/ IL21

This combination has shown great efficiency to induce B cell proliferation & differentiation toward plasmacells and eventually IgG production

Activation via the CD 40 is indeed needed to trigger Ig secretion by B cells, both of naive B cells, mature, or quiescent.

As for the addition of cytokines, CpG, hormones and so on, it depends on what you want to do.

But be aware that the CD40 system is quite sufficient to itself and not require additional signals.

Cross stimulation is obtained with the MAB89 capture, followed by CD154 in its trimeric form (eukaryotic cell surface).

The action of the anti surface IgD and surface IgM, can effectively induce proliferation without any detectable antibody secretion.
