I'm working on a consolidated bioprocess using hemicellulose.
For this, I have to measure cell growth using xylan as a carbon source.
But the xylan containing media is too dark to measure optical density at 600 nm for cell concentration.
Can anyone give me some advice about this?
Your growth analysis depends on microbe you use...if its a yeast then you can go for enumeration using neubauer chamber and can get cell growth result as cells/ml of your broth...if its a bacteria then it creates problem in OD measurements which are not reproducible...to some extent you can try using your media without cell growth as blank and check OD of your broth containing grown cells against that media blank...or else go for plate count enumeration method for bacteria count...
As a simplest approach you can track your cell growth qualitatively by doing wet mount and analysis under optical microscope at 40X or 100X for any increase in cell population/Density visually...Its just to get a rough idea about growth status of your cells...

The method we use consist of recovering the inoculum by centrifugation, followed by washing and resuspending it in water to obtain a clear high cell density suspension.
