I have performed a footshock stress protocol on rats and I am measuring ACTH from plasma. I have already extracted the plasma and it is frozen. I am preparing to run an ELISA on the samples using ACTH kit from Phoenix Pharmaceuticals (http://www.phoenixpeptide.com/catalog/product_info.php?products_id=4203) and it suggests a peptide extraction. Has anyone done this, or not done this? What are the advantages and disadvantages?
Hi Sara. I have experience in determining ACTH in rats' plasma. In my research I use ACTH ELISA kit from Phoenix Pharmac. (used the same cat. EK-001-21, now using fluorescent cat. FEK-001-21). On microplate I add plasma extracted from blood by centrifugation (without dilution because it isn't neccessary). I haven't done peptide extraction and it goes well all the time.
