I isolated phage from river water using Klebsiella pneumoniae.
During spot assay (spot on bacterial lawn), it gave a clear plaque lysis.
However, when I tried plaque assay (mix soft agar with bacteria and phage before pouring onto agar plate), I saw turbid and irregular patches.

Not sure why am I observing this difference? Or am I doing something wrong?

Have you tried repeating it, it might just be some issue with the plates or conditions. But assuming those are real plaques on the left, then you should still see plenty with a 10-fold dilution.
