Is there an easy way to deliver lipids in vitro to crude membrane preparations? The membranes have been prepared from cell lysates by ultracentrifugation in non-denaturing, detergent-free buffer. I would now like to enrich these membranes in vitro with lipids such as cholesterol, free fatty acids or phospholipids. I have done this in the past in intact cells by incubation with lipids complexed by MBCD or bound to serum albumin but am now wondering whether anyone can provide a facile protocol to do this in vitro on purified membranes? Many thanks in advance!
