Normally, if we need to isolate the lymphocytes from the lungs, perfusion through the heart is necessary, by which the blood lymphocyte contamination will be excluded and less red blood cells benefit the flow cytometry.
However, in terms of organ CFU monitoring, I tend to believe that perfusion is unnecessary.

yes, perfusion is unnecessary in organ CFU detection, as RBS contamination is not an issue.
simply you can just get the organ; partially or totally (I usually plate the whole left lung) and directly homogenize it in PBS then you will have a tissue suspension that can be used in dilutions and plating
