I've been using Brij-35 as a component for both wash buffer (1x PBS) and diluent buffers (1x PBS-1%BSA) in my inhibition ELISA experiments. I also use 1x PBS-1%BSA for blocking. Now, whenever I substitute Brij-25 with Tween 20 (0.05%) in both wash and diluent buffers, my relative potencies drop significantly with accompanying T-test failures. The same is observed when switching Brij-35 to Tween 20 in just the wash buffer or just the diluent buffers. What could be some plausible explanation for this decrease in relative potency when using Tween 20?
