Hi, I am having some issue with our rocker. It broke down and it will take time to be fixed. Is there any other solution for me to proceed western blot without rocker? It seems to be impossible. Apparently we only have one rocker since there are not many science postgrad in the department. It will be difficult for me to find replacement at the moment.
Just make sure your membrane is fully covered in the blocking reagent or the antibody solution, you will okay with no rocker. I sometimes used only about 3 ml of antibody solution, and a rocker will instead cause the membrane to dry up during agitation, and therefore, I did not use a rocker. My blots turned out just fine without a rocker.
For the washing step, you can just immerse in PBST multiple times (3 or 5 times) by rocking using your hands for a few seconds until the TBST has a clear texture (i.e. no traces of the leftover milk solution).
Thanks Nurul. I managed to get a normal orbital shaker, not the 3D type from chemistry lab. That should be fine right although it is not like the rocker.
