Anyone who can help me with this question?!
a. Always start culturing the cell line in the quarantine incubator, not sharing the culture medium with your colleagues, and test it for Mycoplasma as soon as possible, regardless of what the person that passed you the cell tells you about its Mycoplasma status.
b. Test for Mycoplasma only if you are told that the line has not been tested for Mycoplasma recently.
c. Grow in your regular incubator without antibiotics initially to let any potential contamination to be easily detected by the turbidity of the medium.
From common knowledge, I would choose 'a', to confirm everything is ok before you use it to do experiments or share it with your co-workers, although I work on plants and plant cells. I would not take other people's words for granted.
You have to check several points.
1) Identity (by testing expression of adequate markers (mRNA AND protein level)
2) potential contamination of cells (bacterial, mykotox, plasmodium, vials/phage)
3) optimal culture conditions (especially for your prefered (adequate) experimental setting, e.g. optimal media and buffers, is our equipment working relyably (conditions CO2, Temp constant), is your stimulus woring in the medium/buffer/working solution. etc.)
