Hello everyone, my interest is to understand the molecular mechanisms involved in anaphylactic shock using Wistar rats. While i inject non-allergic naive rats with iv bolus (1 mg ova in 1 ml), ideally, the BP should decrease for first 2-4 minutes and should get back to baseline levels because the animals are non-allergic. However, in my experiments, the BP decreases and the animal dies after 40 minutes. I was wondering about the cause. Is it because of the volume of IV injection or any other cause. It would be really helpful if you could please suggest on this experiment. Thanks
Dear Richard, with regards to your queries, I think the following response may help to sort out your issues.
1. First of all, I think injection volume might be higher, usually, we use to inject 0.3 or 0.5ml I.V. better use 0.5ml that will work better.
2. 1mg OVA based body-weight might be sensitive to your animal, I suggest try to fix dose less than 1mg (e.g. 0.3, 0.5 or 0.7mg/kg b.w) by analyzing BP levels.
3. Since a non-allergic animal, analyzing cytokine levels by simple ELISA is not a good option, alternatively, you can lung morphology because OVA induces sudden respiratory (asthma) effects. I hope it may help.
Dear Mani, thanks a lot for your suggestions. This model worked previously in the lab so ova dose is published already. however, i agree with the volume of saline injection. i will repeat the experiment with lower volume and let you know the outcome.
