I wish to silanize gold substrate to immobilized my proteins for SPR studies. Can anyone suggest a method to generate hydroxyl group. can we go for oxygen plasma or air plasma? will it affect SPR response?
If you want to immobilize proteins on the gold substrate then you do not need neither silanization nor hydroxilation. What you need is to coat the gold substrate with self assembled monolayer of carboxyl-functionalized thiols - for instance with mercaptoundecanoic acid (MUA). Then you can covalently immobilize proteins using EDC reaction. Such immobilzation strategy is very well known and described.
In general, cleaning of the surface is necessary for a succesfull immobilization.Then,it needs to thiolate the gold substrate and may follow by several experimental steps which are designed according to targeted functional groups.
