TE/TW solution:

Add 10mM Tris pH 8.0 + 1mM EDTA

and to the resultant solution add 0.01% Tween-­‐20

1) Prepare 100 mL of 10 mM Tris and 1 mM EDTA. For example:

- take 100 mL of Milli-Q or deionized water

- add 121.1 mg of Tris (MW=121.14, Sigma-Aldrich 252859)

- add 29.2 mg of EDTA (MW=292.24, Sigma-Aldrich E9884)

2) Check the pH and adjust to what you need with HCl/KOH (range: 7.0-9.0).

2) 1% means 1 mL for 100 mL. So, for 0.01% Tween, add 10 uL of Tween-20stock into your 100 mL of buffer made in 1).

Phosphate-buffered saline (PBS) containing 0.1% (v/v) Tween 80 (PBS-T). Diluting and washing reagent; add 2.0 mL of polysorbate 80 (Tween 80) to 200 mL PBS (10X) solution. Mix thoroughly and (using a volumetric flask) bring solution to volume (2 L) with deionized water. Distribute into bottles and filter sterilize.
