Most of the researchers are following MS medium, formulated by Murashige and Skoog. As it is new plant, you can try for Modified MS medium also (MS salts + B5 vitamins). Cooncentrations and combinations of plant growth regulators depend on the type of explant. Initially you try for shoot tip explant. So try for Direct organogenesis. Standardization of protocol plays a major role in tissue culture. Lots of research papers available for the hormone concentrations, you can try it for that with little modifications. So you have to optimize the protocol for the new plant. Because several factors plays a role eg. age of the explant, type of explant, culture conditions, pH of the medium, concentrations of growth regulators etc.
Tissue culture is a general term and there are several kinds and objectives of tissue culture. For example, it may be for tissue regeneration, haploid formation, plant regeneration, etc. Accordingly, protocols also differ. Tissue culture is generally affected by the type of explant used, culture media, pretreatments, incubation conditions, etc. Therefore, IF YOU MENTION SPECIFIC OBJECTIVE, I MAY SUPPLY YOU DETAILED AND FULL PROOF PROTOCOLS.
Plant is impatiens...on impatiens there is no authentic work available i search alot..and this plant is ornamental...i want to produce virus free plant through tissue culture. And through tissue culture to improve for secondary metabolite as well.
