what could be the possible reason for my transformed cells to not express proteins
Sometimes it happens when plasmid is ligated without insert, Usually I check my insert in construct by restriction analysis. Or as was said early you have a wrong context.
If all is correct with plasmid, you can check your protein by western blot, I had situation, when there was no overexpression, in this case I try to found the best conditions. Sometimes a changing a tag or position of tag helps
Only carbenicillin selection is not enough, you have to make sure that the plasmid is transformed in DE3 cells using other mol bio methods. Once you are sure that the cells got the gene, you can do multiple manipulation to get higher expression level but if still you don't see expression it is better to use other strains for exprssion.
