This is common phenomenon in most cell differentiation protocol. You have to be very careful while changing the media as they loos binding from the substratum. You should change one well at the one time and then go for next. This is time consuming but worked for me.
If possible you can use coated plates, now a days they are commercially available but they are costly. Or you can coat by yourself with poly-l-lysin.
The main issue is when i seeds them with coverlips, after the 4th day of differentiation they start to peel off the glass. I´ve tried really carefully and slowly when it comes to the medium change but still occurs. Any idea why ?
