I am using M13 tailed primers with the forward which initially starts the amplification PCR reaction. When the part of the Forward Primer is cut off, can the M13 tailed primer amplify later on substituting the forward primer? I need some help on this matter because in theory it might work because the M13 which is combined with the Forward primer in the beginning has been sequenced according to the DNA template, and the cut part template is the same DNA template for a second time.
Hi Aldosky, really this is theoretically correct but practically if there is a cut off in the froward primer this will be affect PCR reaction and also the final product as the aim from M13 tailed primer is to facilitate and specify the sequence process of the amplified product.

I think Dr.Ehab on right but the defect will depend on the length of the tail.
