Without the addition of PHA, karyotyping (finding metaphases) can only be performed in bone marrow. When peripheral blood (lymphocytes) is used, no metaphases will be found without PHA.

Without PHA there will be no metaphases in peripheral blood cultures. A new blood sample should be taken and PHA added.
