Hi Lleretny, we use in our experiments HBM Exosome RNA extraction kit, it provides immunobeads for overall exosomes capture, all the solutions you need and also columns and elution tubes. Kit allows to extract total RNA, including miRNA. The catalog number is HBM-BOF-10(20)-#. Good luck!
We use TRIzol® RNA Isolation Reagents (invitrogen, Catalog number: 15596026) to extract total RNA from exosomes and cells.
Stem-loop qRT-PCR is used to validate miRNA expression levels in exosomes and cells. iScript Reverse Transcription Supermix for RT-qPCR (BIO-RAD, Catalog number: 1708841) is used to synthesize cDNA transcripts using RT-primers and SsoAdvanced™ Universal SYBR® Green Supermix (BIO-RAD, Catalog number: 1725275) is used for real-time PCR.
The qRT-PCR is run on a CFX96 Touch™ Real-Time PCR Detection System (also from BIO-RAD).
