I am using 1/50 dilution of (50-250 microgram concentration) and 3 ml of H2O2 but absorbance of sample continuously increasing. Or some time negative absorbance also getting I don't understand what is the result behind it.

There are many issues that you need to check.
1-did you use quartz cell?
2- did you use dry or fresh sample?
however, the negative absorbance indicates that the sample does not contain enough enzyme that mean that you need to decrease the dilution ratio
To be sure that there is an activity you should try different volumes of the diluted as well as of the undiluted extract.
