Have you diluted your samples? If yes, try using them undiluted. Otherwise your samples might contain very low concentrations of IL-18 to begin with, try concentrating your samples.

The samples are undiluted. In addition to serum/plasma, I already try cell culture supernants, from human macrophages stimulated with NLRP3 inflammasome agonist.

Since your controls are working, I'd suggest contacting the company where you got your ELISA from, if it is technical issue they will help you but if not then I think concentrating your samples is the best bet.
