The molecular weight of Tris and KCl should be on the bottle/container.

Molecular weight X desired concentration x volume = amount of chemical you'll need.

Assuming molecular weight of KCl 74.55 g/mol

74.55 g/mol X 0.27M X 500 ml =10.06 g

Do the same for Tris.

Calculate amount of glucose

Add into beaker with about 400 ml H2O and disolve

Adjust the pH.

Adjust the volume to exactly 500 ml.
