What is the mechanism of CD3 Ab alone to activate T cells? It is different from antigen dependent activation which requires MHC molecules. It seems these are separated mechamisms, but also closely related to each other. I would like to understand more details.
The antibody targets the signaling portion of the TCR, which is CD3. The "difference" is that for MHC presented peptides, the TCR will have to recognize the peptide presented by the MHC, and this "recognition" activates the CD3 to signal. The antibody bypass the alphabeta or the gammadelta regions of the TCR.
As for what type of T cells thta CD3 activates, everything.
i agree with Shen-An. Practically: anti-CD3 can activate T cells alone and without IL-2 they will do AICD: activation induced cell death.
with (moderate to high amounts) IL-2 you will get a strong proliferation. These have been called LAK cells, leukocyte artifical killer... because they loose the MHC restriction. As far as i know, it is not clear whether those come from a certain population and expand (was once believed a source for NK cells, NKT etc. but doesnt matter), or whether normal T cells get changed...

41-BB and/or CD28 stimulation can cause non-MHC-restricted activation of T-cells. It is used in creation of CAR T-cells for treatment of malignancies
