I am having trouble with a cytotoxic assay :

I plate a certain cell number in 96w plates. On the next day, I add serially diluted antibodies on the well and use AlamarBlue as a readout for cytotoxicity 96h later (confluence in the vehicle well is of 80%).

I see a cytotoxicity activity of Trastuzumab in the nanomolar range on a cell line that is HER2-negative (negative control). After having tried various sources of Trastuzumab and various HER2-negative cells, I am puzzled on how to troubleshoot this assay.

Any idea ?
