Its an obsolete term which is abandoned and not in use anymore. Its use is just wrong now.

To isolate enzymes, first you need to know what are your candidates and then there are several techniques which can be used. Basically they are protein isolation methods and their separation can be done by PAGE or 2D electrophoresis. These are just for the characterization or finding out what your candidates are. Later for the confirmation, sequencing can be performed or other assays like ELISA etc.
