The difference between PEG400 and PEG600 for Transformation applications in Haloferax volcanii?

Dear all,

I'm currently working with halophilic archaea Haloferax volcanii. I'm going to try the transformation with using a spheroplasting protocol and PEG (Halohand Book).

The original protocol use 60% PEG600, but we have only PEG400. Is it significant to use exactly PEG600? Or the PEG400 is enough?

Using PEG400 for your application should be fine. There is not really that much difference between PEG400 and 600, the population of molecular sizes between those two are significantly overlapping.
