Getting agrobacterium growth on my resistant callus repeated times, even washing 4-5 times with 200mg/l timetin, can anyone suggest .. how to get rid of this probblem?
1. Press your infected callus explants in 3-4 layers of sterilized filter papers in petri plate (or repeat this step) to blot the excess Agrobacterium culture.
2. Then dried explants rinsed in sterile distilled water for 3-4 times and inoculated on respective medium supplemented with your antibiotic.
3. Sub-culture explants after every two weeks on same media.
Try to used broad spectrum anti biotic to kill bacteria on repeated basis as your tissue is callus where cells are tightly packed. Rest follow jaspreet on weekly or wi weekly transfers.
Rishi Kesh Agrobacterium is a bacterium easy developing antibiotic resistance. Isolate bacteria from the callus and test antibiotic tolerance using standard disk method. Large number of commercial antibiotics on the disks is available for medical purposes. Efficient antibiotics must be tested on plants for their phytotoxic effect.
