I'm a student and it's getting common to have smileys in the dna ladder. We use molecular weight marker viii (Roche), ~100 - 1114bp. 100V at 1 hour. Same results at 80V for 1,5 hour. 4ul ladder, 16ul dna, 5ul orange g. Added gelred prior to hardening of the gel
The 1x tbe buffer is used several times by several students, it would be no surprise if the ladder is old.
How am I supposed to define the fragment sizes? If I aim for the top of the smileys, my fragments would be kind of the right size, but with massive guesses since the ladder smears. But if I use the bottom of the smileys, every sample would have fragments too big.
